Background

The CRISPR/Cas9 knockout system is composed of a Cas9 protein with DNA cleavage activity and a gRNA that recognizes a specific target. This simplifies the construction of knockout vectors because target recognition can be achieved by editing only the gRNA in the system. In mammalian cell culture systems, most gene editing is accomplished by transient transfection and lentiviral transduction, which is effective for routine low-throughput applications. However, for other applications, it may be more effective to stably express one of the components, such as the CRISPR-Cas9 nuclease.

It is important to note that there are differences in the efficiency of individual sgRNAs themselves. Prior to extensive screening efforts, it is necessary to determine which CRISPR sgRNAs are more efficient at genome editing. Researchers can assess the efficiency and specificity of Cas9-mediated genome editing by introducing sgRNAs into Cas9-stabilized cell lines.

  • Gene knockout using lentiviral CRISPR

Cas9 stable cell lines are useful for gene knockout experiments using the lentiviral CRISPR system. Transduction of Cas9 stable cell lines with lentivirus expressing sgRNA enables efficient genome editing.
