Introduction of CRISPRi

The CRISPRi system is based on CRISPR/Cas9, which can specifically inhibit the transcription of target genes rather than knockout the genes to achieve regulation. CRISPRi can knock out target genes at different levels and inhibit multiple genes simultaneously with reversible effects. The technology uses dCas9 as a synthetic transcription factor, which is a mutant Cas9 protein that lacks nuclease cleavage activity and produces programmable RNA-guided DNA-binding proteins in human cells. dCas9 is associated with transcriptional repressors, such as KRAB (Kruppel-associated box). dCas9-KRAB fusion proteins bind to the transcription start site (TSS) of target genes under the guidance of gRNA, blocking RNA polymerase and transcript elongation, and silencing the expression of target genes.

To construct CRISPRi cell lines, we integrate dCas9 (catalytically inactivated Cas9) proteins into stable host cell lines to ensure consistent and heritable gene regulation. We select commonly used and well-characterized host cell lines such as blood lineage cells, cancer cell lines, stem cells (iPSC) or other relevant cell lines to meet the research demands of different clients.
