Since its inception, CRISPR/Cas9 technology has rapidly evolved, becoming a hot topic in biological research. On November 16, 2023, the Medicines and Healthcare Products Regulatory Agency (MHRA) granted conditional marketing authorization for the gene therapy CASGEVY. CRISPR technology relies on single guide RNA (sgRNA) to direct the Cas9 protein to the target DNA sequence, achieving precise gene editing. The sgRNA, with its 20nucleotide guide sequence, pairs with the target DNA sequence, ensuring Cas9 cuts the DNA at the correct position. This guiding function not only determines the specificity and efficiency of the CRISPR system but also allows optimization of the sgRNA sequence to reduce off-target effects, enhancing the accuracy of gene editing.
