Introduction of CRISPRa

CRISPRa, also known as CRISPR activation, works by binding dCas9 to various transcriptional activators to increase the expression of target genes. dCas9 can activate endogenous promoters for efficient transcription without the need for additional exogenous expression elements. It is not limited by gene transcript size. Compared to traditional overexpression techniques, CRISPRa promotes gene expression by activating endogenous promoters for efficient transcription without the need to construct additional exogenous expression elements. Additionally, it is not limited by the size of the gene transcript. dCas9-mediated gene regulation is reversible and does not permanently modify the genomic DNA. Furthermore, it is possible to design multiple gRNAs to regulate multiple target genes simultaneously. Additionally, fusing dCas9 with fluorescent proteins enables tracking of target gene location.

Overview of Our CRISPRa Cell Lines

To create CRISPRa cell lines, we combined catalytically inactivated Cas9 (dCas9) proteins with transcriptional activators. This was done to precisely activate the target gene in the host cell line. We then integrated this fusion construct into the genome of the target cell line using a lentiviral transduction approach. To generate a stable CRISPRa cell population with the correct protein expression levels, we used advanced screening techniques to obtain a monoclonal cell population. We then tested the CRISPRa activity using a functional assay. Our high-quality CRISPRa cell lines provide reliable and reproducible results for our clients, enabling researchers to achieve innovative results in the field of gene regulation.
