Background

Applications

NECTIN4, also known as nectin cell adhesion molecule 4, is a gene encoding a member of the nectin family of cell adhesion molecules. NECTIN4 is primarily expressed in epithelial tissues, including the skin, lungs, and bladder, and is involved in cell-cell adhesion, migration, and proliferation. Nectin-4 protein is initially synthesized as a precursor and undergoes glycosylation and other post-translational modifications to become the mature form. The mature nectin-4 protein is then localized to the cell membrane, where it interacts with other cellular proteins to mediate its actions. One of its key roles is to promote the formation of homo- and heterodimers with other nectin family members, such as NECTIN1, NECTIN2, and NECTIN3, to form functional adhesion complexes. These complexes are essential for the formation of a tight barrier between epithelial cells and play a role in signaling pathways that regulate cell behavior. In terms of disease, abnormal expression of NECTIN4 has been associated with multiple types of cancer, including breast, ovarian, and bladder cancers. Due to its restricted expression in normal tissues but prominent in certain cancers, NECTIN4 has emerged as a potential target for cancer therapy. In addition to its role in cancer, NECTIN4 has been implicated in a genetic disorder called ectodermal dysplasia-syndactyly syndrome type 1 (EDSS1), which is characterized by abnormalities of the skin, hair, teeth, and nails, indicating the importance of the normal function of NECTIN4 in normal human development.
Cancer Research: NECTIN4 is overexpressed in a variety of cancers, including breast, ovarian, and lung cancers. The human NECTIN4 knockout cell line, HEK293T, allows for in-depth studies of the role of NECTIN4 in tumorigenesis and cancer progression. Drug Development: By utilizing the NECTIN4 knockout cell line, pharmaceutical researchers can test the efficacy and specificity of drugs targeting NECTIN4. This helps identify potential therapeutic compounds and assess their effects on cellular processes. Immune Response Studies: NECTIN4 has been shown to play a role in immune cell interactions. Knockout cell lines can be used to study how NECTIN4 loss affects immune cell adhesion, migration, and signaling. These insights can help understand immune evasion mechanisms in cancer and develop immunotherapies. Cell Signaling Pathways: NECTIN4 is part of the nectin family and is involved in various cell signaling pathways. Studies utilizing the NECTIN4 knockout cell line can elucidate specific signaling pathways in which NECTIN4 participates. Developmental Biology: NECTIN4 is involved in the formation and maintenance of adherens junctions. NECTIN4 knockout cell lines can be used to study how NECTIN4 loss affects cell structure and tissue development.
